The first appearance of Spider-Man was in AMAZING FANTASY #15, which had a date of August 1962. We won't go into all the details here, but Spider-Man is said to have been around in some form for some years before that. Here is a story from 1960 with "Spider Men" by Jack Kirby, one of the people who helped to develop the character of Spider-Man.

This story came out in 1960, which was a little before Spider-Man, and a little after the movie EARTH VERSUS THE SPIDER. There are a number of similarities between the two, including the use of DDT. Some of the panels in the story even resemble scenes from the movie.
The 2001 EARTH VERSUS THE SPIDER reused the title, but was not actually a remake. It was described by co-creator Stan Winston as being about, "a young man whose attempt to become a Spider-Man type superhero goes disastrously wrong, leaving him with a Jekyll-and-Hyde type personality disorder".
Which sounds like it had about as much to do with the comic books as it did with the previous movie.
